They are serine protease inhibitors and belong to the serpin superfamily of proteins, which utilize a unique conformational change mechanism for protease inactivation. Antithrombin is a vital plasma anticoagulant due to its ability to inhibit blood-clotting proteases. Protein C inhibitor PCI ; is believed to be the most important regulator of the protein C system of blood coagulation, by acting as an inhibitor of the anticoagulant activated protein C APC ; in plasma. Both these inhibitors have been found to possess additional functional properties. Antithrombin has antiinflammatory, antiangiogenic and antitumor properties in addition to acting as an anticoagulant, whereas PCI, in addition to inhibiting APC, is capable of inhibiting several other serine proteases involved in blood coagulation, fibrinolysis, fertilization and degradation of components of the extracellular matrix. Antithrombin and protein C inhibitor are N-glycosylated with four and three potential N-glycosylation sites, respectively. They both have the ability to bind to glycosaminoglycans, which thereby regulate their inhibitory activities towards the target proteases receptors. PCI has additionally been found to interact with certain lipids, such as retinoic acid and oxidized phospholipids. The aim of this project is thus to study the multifunctionality of the inhibitors and the specific objectives are to study the roles of: i ; N-linked glycans attached to the inhibitors ii ; interactions with glycosaminoglycans iii ; interactions with hydrophobic compounds on regulating the different functions activities exerted by the two serpins.
